Yeah, it could.
To work, the key fob needs to be recognized by the module, and if the battery is dead, then the module is simply not powered anymore. At this point, whether the key fob's battery is ok or not, if the module doesn't respond, nothing will happen.
Kinda like trying to use a phone with a good battery while the cell phone tower is down — the signal goes out of the key fob fine, but there's nothing to receive it.
And yes, I understand that the problem is then that you first need to unlock the doors so you can then pop the hood and charge/replace your battery, which you can't do with an inoperative key fob. Luckily, all manufacturers think of a procedure to open the doors with a physical key in such a condition, which you will find in this 2004 Chevrolet Avalanche Owners Manual.
